    The implementation of the HAL/S language on the IBM-360, and in particular the mechanization of its real time, I/O, and error control statements within the OS-360 environment is described. The objectives are twofold: (1) An analysis and general description of HAL/S real time, I/O, and error control statements and the structure required to mechanize these statements. The emphasis is on describing the logical functions performed upon execution of each HAL statement rather than defining whether it is accomplished by the compiler or operating system. (2) An identification of the OS-360 facilities required during execution of HAL/S code as implemented for the current HAL/S-360 compiler; and an evaluation of the aspects involved with interfacing HAL/S with the SUMC operating system utilizing either the HAL/S-360 compiler or by designing a new HAL/S-SUMC compiler

    The relationship between substance use disorders and psychiatric pathology is still an open question. The main aim of the present study was to verify whether the five psychopathological dimensions identified through the SCL-90 tool in a previous study carried out on patients with heroin addiction entering an outpatient opioid agonist treatment (OAT) were also observable in those entering a residential treatment community (TC). Further aims were to look at differences in the psychopathological profiles of patients entering a TC versus an OAT treatment and at the correlation between gender and the observed psychopathology.A confirmatory factor analysis was performed on the results of SCL-90 filled by 1,195 patients with heroin dependence entering TC treatment. It replicates the extraction method previously used on 1,055 OAT patients with heroin addiction by using a principal component factor analysis (PCA). The association between the kind of treatment received (TC or OAT), gender, and the psychopathological dimensions was assessed through logistic regression and general linear model (GLM) analysis.The PCA carried out on the SCL-90 results of patients entering a TC yielded a five-factor solution, confirming the same dimensions observed in patients entering an OAT: 'worthlessness and being trapped', 'somatization', 'sensitivity-psychoticism', 'panic anxiety', and 'violence-suicide'. The logistic regression analysis showed a statistically significant association between 'somatization' and 'violence-suicide' severity score and OAT. GLM analysis showed that psychopathological factorial scores for 'worthlessness-being trapped', 'somatic symptoms', and 'panic anxiety' dimensions were more severe in OAT vs TC male patients and in TC vs OAT female ones. 'Violence suicide' followed the same severity pattern for males, but did not differ in TC vs OAT females, while 'sensitivity-psychoticism' did not differ in OAT vs TC patients. The five dimensions did not differ in OAT males vs females.Our research appears to confirm the existence of a specific aggregation of psychological/psychiatric features within the category of individuals with heroin addiction. It also shows a correlation between the dominant psychopathological subgroup and the assignment to TC versus OAT. Further research is needed to clarify the differences between the five psychopathological subgroups and their determinants

    Psychopathology of addiction: May a SCL-90-based five dimensions structure be applied irrespectively of the involved drug?

    BACKGROUND: We previously found a five cluster of psychological symptoms in heroin use disorder (HUD) patients: ‘worthlessness-being trapped’, ‘somatic-symptoms’, ‘sensitivity-psychoticism’, ‘panic-anxiety’, and ‘violence-suicide’. We demonstrated that this aggregation is independent of the chosen treatment, of intoxication status and of the presence of psychiatric problems. METHODS: 2314 Subjects, with alcohol, heroin or cocaine dependence were assigned to one of the five clusters. Differences between patients dependent on alcohol, heroin and cocaine in the frequency of the five clusters and in their severity were analysed. The association between the secondary abuse of alcohol and cocaine and the five clusters was also considered in the subsample of HUD patients. RESULTS: We confirmed a positive association of the ‘somatic symptoms’ dimension with the condition of heroin versus cocaine dependence and of the ‘sensitivity-psychoticism’ dimension with the condition of alcohol versus heroin dependence. ‘Somatic symptoms’ and ‘panic anxiety’ successfully discriminated between patients as being alcohol, heroin or cocaine dependents. Looking at the subsample of heroin dependents, no significant differences were observed. CONCLUSIONS: The available evidence coming from our results, taken as a whole, seems to support the extension of the psychopathological structure previously observed in opioid addicts to the population of alcohol and cocaine dependents

    Spatial and seasonal variations of aerosols over China from two decades of multi-satellite observations – Part 2: AOD time series for 1995–2017 combined from ATSR ADV and MODIS C6.1 and AOD tendency estimations

    Understanding long-term variations in aerosol loading is essential for evaluating the health and climate effects of airborne particulates as well as the effectiveness of pollution control policies. The expected satellite lifetime is about 10 to 15 years. Therefore, to study the variations of atmospheric constituents over longer periods information from different satellites must be utilized.Here we introduce a method to construct a combined annual and seasonal long time series of AOD at 550 nm using the Along-Track Scanning Radiometers (ATSR: ATSR-2 and AATSR combined) and the MODerate resolution Imaging Spectroradiometer on Terra (MODIS/Terra), which together cover the 1995–2017 period. The long-term (1995–2017) combined AOD time series are presented for all of mainland China, for southeastern (SE) China and for 10 selected regions in China. Linear regression was applied to the combined AOD time series constructed for individual L3 (1°&thinsp; × &thinsp;1°) pixels to estimate the AOD tendencies for two periods: 1995–2006 (P1) and 2011–2017 (P2), with respect to the changes in the emission reduction policies in China.During P1, the annually averaged AOD increased by 0.006 (or 2&thinsp;% of the AOD averaged over the corresponding period) per year across all of mainland China, reflecting increasing emissions due to rapid economic development. In SE China, the annual AOD positive tendency in 1995–2006 was 0.014 (3&thinsp;%) per year, reaching maxima (0.020, or 4&thinsp;%, per year) in Shanghai and the Pearl River Delta regions. After 2011, during P2, AOD tendencies reversed across most of China with the annually averaged AOD decreasing by −0.015 (−6&thinsp;%) per year in response to the effective reduction of the anthropogenic emissions of primary aerosols, SO2 and NOx. The strongest AOD decreases were observed in the Chengdu (−0.045, or −8&thinsp;%, per year) and Zhengzhou (−0.046, or −9&thinsp;%, per year) areas, while over the North China Plain and coastal areas the AOD decrease was lower than −0.03 (approximately −6&thinsp;%) per year. In the less populated areas the AOD decrease was small.The AOD tendency varied by both season and region. The increase in the annually averaged AOD during P1 was mainly due to an increase in summer and autumn in SE China (0.020, or 4&thinsp;%, and 0.016, or 4&thinsp;%, per year, respectively), while during winter and spring the AOD actually decreased over most of China. The AOD negative tendencies during the 2011–2017 period were larger in summer than in other seasons over the whole of China (ca. −0.021, or −7&thinsp;%, per year) and over SE China (ca. −0.048, or −9&thinsp;%, per year).The long-term AOD variations presented here show a gradual decrease in the AOD after 2011 with an average reduction of 30&thinsp;%–50&thinsp;% between 2011 and 2017. The effect is more visible in the highly populated and industrialized regions in SE China, as expected.</p

    Tumour-suppression function of KLF12 through regulation of anoikis

    Suppression of detachment-induced cell death, known as anoikis, is an essential step for cancer metastasis to occur. We report here that expression of KLF12, a member of the Kruppel-like family of transcription factors, is downregulated in lung cancer cell lines that have been selected to grow in the absence of cell adhesion. Knockdown of KLF12 in parental cells results in decreased apoptosis following cell detachment from matrix. KLF12 regulates anoikis by promoting the cell cycle transition through S phase and therefore cell proliferation. Reduced expression levels of KLF12 results in increased ability of lung cancer cells to form tumours in vivo and is associated with poorer survival in lung cancer patients. We therefore identify KLF12 as a novel metastasis-suppressor gene whose loss of function is associated with anoikis resistance through control of the cell cycle

    Assessment of Arthrobacter viscosus as reactive medium for forming permeable reactive biobarrier applied to PAHs remediation

    Polycyclic aromatic hydrocarbons (PAHs) are significant environmental contaminants as they are present naturally as well as anthropogenically in soil, air and water. In spite of their low solubility, PAHs are spread to the environment, and they are present in surface water, industrial effluent or groundwater. Amongst all remediation technologies for treating groundwater contaminated with PAHs, the use of a permeable reactive biobarrier (PRBB) appears to be the most cost-effective, energy efficient, and environmentally sound approach. In this technology, the microorganisms are used as reactive medium to degrade or stabilize the contaminants. The main limits of this approach are that the microorganisms or consortium used for forming the PRBB should show adequate characteristics. They must be retained in the barrier-forming biofilm, and they should also have degradative ability for the target pollutants. The aim of the present work is to evaluate the viability of Arthrobacter viscosus as bioreactive medium for forming PRBB. Initially, the ability of A. viscosus to remove PAHs, benzo[a]anthracene 100 μM and phenanthrene 100 μM was evaluated operating in a batch bench-scale bioreactor. In both cases, total benzo[a]anthracene and phenanthrene removals were obtained after 7 and 3 days, respectively. Furthermore, the viability of the microorganisms was evaluated in the presence of chromium in a continuous mode. As a final point, the adhesion of A. viscosus to sepiolite forming a bioreactive material to build PRBB was demonstrated. In view of the attained results, it can be concluded that A. viscosus could be a suitable microorganism to form a bioreactive medium for PAHs remediation.This work has been supported by the Spanish Ministry of Economy and Competitiveness and FEDER Funds (Project CTM 2011-25389).     Transcription-replication conflicts: How they occur and how they are resolved

    The frequent occurrence of transcription and DNA replication in cells results in many encounters, and thus conflicts, between the transcription and replication machineries. These conflicts constitute a major intrinsic source of genome instability, which is a hallmark of cancer cells. How the replication machinery progresses along a DNA molecule occupied by an RNA polymerase is an old question. Here we review recent data on the biological relevance of transcription-replication conflicts, and the factors and mechanisms that are involved in either preventing or resolving them, mainly in eukaryotes. On the basis of these data, we provide our current view of how transcription can generate obstacles to replication, including torsional stress and non-B DNA structures, and of the different cellular processes that have evolved to solve them

    The SUMO Isopeptidase Ulp2p Is Required to Prevent Recombination-Induced Chromosome Segregation Lethality following DNA Replication Stress

    SUMO conjugation is a key regulator of the cellular response to DNA replication stress, acting in part to control recombination at stalled DNA replication forks. Here we examine recombination-related phenotypes in yeast mutants defective for the SUMO de-conjugating/chain-editing enzyme Ulp2p. We find that spontaneous recombination is elevated in ulp2 strains and that recombination DNA repair is essential for ulp2 survival. In contrast to other SUMO pathway mutants, however, the frequency of spontaneous chromosome rearrangements is markedly reduced in ulp2 strains, and some types of rearrangements arising through recombination can apparently not be tolerated. In investigating the basis for this, we find DNA repair foci do not disassemble in ulp2 cells during recovery from the replication fork-blocking drug methyl methanesulfonate (MMS), corresponding with an accumulation of X-shaped recombination intermediates. ulp2 cells satisfy the DNA damage checkpoint during MMS recovery and commit to chromosome segregation with similar kinetics to wild-type cells. However, sister chromatids fail to disjoin, resulting in abortive chromosome segregation and cell lethality. This chromosome segregation defect can be rescued by overproducing the anti-recombinase Srs2p, indicating that recombination plays an underlying causal role in blocking chromatid separation. Overall, our results are consistent with a role for Ulp2p in preventing the formation of DNA lesions that must be repaired through recombination. At the same time, Ulp2p is also required to either suppress or resolve recombination-induced attachments between sister chromatids. These opposing defects may synergize to greatly increase the toxicity of DNA replication stress
